Workflow

How It Works: AquaSpark® Detection Technology

The patented AquaSpark® system detects phosphatidylinositol-specific phospholipase C (PI-PLC), a hallmark virulence enzyme of L. monocytogenes, making this a highly specific detection method. A proprietary mixture of bacteriophages and selective agents in the enrichment broth suppresses competing microflora while selectively promoting recovery of sub-lethally injured L. monocytogenes during the 24-hour incubation period.

Simple 6-Step Workflow

24-hour
  1. Preparation - Wet swab with buffer or neutralizer. Mark/label the swab tubes and the test tubes according to your test plan. Use N-Light™ MaxiSamplers for large or irregular surfaces.
  2. Sample - Swab a 10x10 cm area with firm pressure, rotating the swab in between passes. Apply gentle pressure to penetrate biofilms.
  3. Transfer - Open test tube. Break swab tip into enrichment broth and discard handle. Replace biosafety cap and push down firmly until you hear a click. Briefly shake tube.
  4. Incubate - Place tube in NEMIS Dry Block Heater at 37°C for 24 hours (+/- 1 hour). Do not use a standard microbiological incubator.
  5. Activate - Release AquaSpark® tablet by pressing cap button. Verify tablet drops into liquid. Shake tube 5 seconds to dissolve. Return tube dry block heater at 37°C for 5 minutes.
  6. Measure - Insert tube into NEMIS Bench Top Luminometer exactly 5-7 minutes after tablet release. Measurement takes 10 seconds. Result: >20,000 RLU = Presumptive Positive.

Positive L. monocytogenes environmental monitoring results may trigger corrective action documentation requirements for facilities subject to Preventive Controls for Human Food. Samples should be taken according to your HACCP plan, examples include from production lines (pre- and post-production), food contact surfaces, etc. N-Light™ Listeria monocytogenes is compatible with environmental monitoring programs designed to support compliance with food safety schemes. Audit-ready data is exportable via the NEMIS data app in CSV/LIMS-compatible format.

Recommendation: Use the N-Light™ MaxiSampler for broad-zone floor and surface surveillance. Always handle post-incubation tubes as potentially infectious materials. Confirmation procedures must be performed in a laboratory. Do NOT dispose of enrichment broth in the sink. Technical FAQ

The N-Light™ L. monocytogenes test is a qualitative test for rapidly detecting the foodborne bacterial pathogen Listeria monocytogenes. It is intended for use in food processing areas and equipment as part of an environmental monitoring program. Being intuitive, rapid, cost-effective, and lab-free, it offers an affordable solution to scale testing and empowers your quality management process.

Yes, the N-Light™ L. monocytogenes has been certified in the AOAC® Research Institute Performance Tested Methods℠ Program (Certificate No. 122002) and validated against the reference method ISO 11290-1:2017.

The only required equipment is the NEMIS dry block heater and theNEMIS Luminometer. They can be used on-site without any laboratory infrastructure. It is recommended to perform the test in a separate room, distant from the production environment.

The N-Light™ L. monocytogenes test broth contains a mixture of nutrients (for bacterial growth) combined with selective factors (limiting the growth of non-target bacteria) such as antibiotics and NEMIS proprietary combination of bacteriophages. This combination creates an ideal culture medium for Listeria monocytogenes leading to superior sensitivity and specificity of the test. Its unique formulation inhibits the growth of competing bacteria and makes the test more reliable in challenging environments. Additional data about the N-Light™ enrichment broth can be found here.

Results can be obtained after 24 hours of incubation at 37°C.  Shortened incubation time less than 23 hours is not AOAC-validated and not encouraged. On the other hand, tubes can be incubated for up to 72 hours.

Hot spots that accurately indicate potential product contamination should be preferentially sampled. Please refer to your Hazard Analysis and Critical Control Points (HACCP) plan.

It is possible to collect samples from production lines or sites that have not been recently cleaned. Sample location and application of N-Light™ L. monocytogenes should be categorized as follows:

  • Clean Surfaces
    The AOAC has validated the full performance of N-Light™ L. monocytogenes
  • Food or dirt residue-exposed surfaces within the production area or before cleaning.
    Samples taken from production lines (both during and after production) and other places in the facility that are regularly cleaned are well within the capacity of this test. Make sure not to overload the sample with food residues. Rather, go into niches and cavities of machines and apply pressure on surfaces.
    These techniques are outside of the scope of the PTM validation performed for this product.
  • Direct sampling of food or in places of high dirt / food residue concentration
    Meticulous sampling is required to ensure no test tube overload. Do not directly collect food residues. Examples of high load sampling spots are uncleaned drains, smeared water tanks, or the inside of uncleaned food residue collection containers.
    These techniques are outside of the scope of the PTM validation performed for this product.

We recommend avoiding sample locations with overload potential and selecting alternative sampling sites in the immediate vicinity when appropriate. If you have questions about sampling site selection, consult your facility's Quality Assurance or HACCP team, as sampling requirements should be based on your site's specific processes, risk assessments, and environmental monitoring program.

Optimally, high-risk spots should be swabbed on a daily basis after each cleaning or production shift. In case of a positive result, consult your facility's HACCP and implement immediate, corrective actions. Lower-risk control points may be tested less frequently. Consult your facility's HACCP.
